donafenib + sintilimab

Donafenib + sintilimab is a combination therapy consisting of donafenib, an oral small molecule multikinase inhibitor targeting multiple kinases involved in tumor angiogenesis and proliferation (notably VEGFR, PDGFR, RAF kinases), and sintilimab, a monoclonal antibody immune checkpoint inhibitor targeting programmed death 1 (PD-1) receptor. This combination is being investigated primarily for the treatment of hepatocellular carcinoma (HCC), including advanced/unresectable HCC and as neoadjuvant or adjuvant therapy in resectable cases. The rationale for combining these agents is to leverage the antiangiogenic effects of donafenib with the immunomodulatory activity of sintilimab to enhance antitumor efficacy. Clinical studies have shown promising results in terms of progression-free survival and overall response rates with an acceptable safety profile[6][9].
