donepezil + choline

Donepezil + choline is a combination therapy consisting of donepezil hydrochloride, a reversible acetylcholinesterase inhibitor, and choline, a precursor to the neurotransmitter acetylcholine. This treatment is being investigated by Sheba Medical Center for the management of autism spectrum disorder (ASD) in children and adolescents. The rationale for this combination is to enhance cholinergic neurotransmission, which is hypothesized to be deficient or dysfunctional in individuals with ASD. Donepezil works by preventing the breakdown of acetylcholine in the synaptic cleft, while choline supplementation provides the necessary substrate for increased acetylcholine synthesis. Clinical evaluation in a Phase 4 trial at the Weinberg Child Development Center demonstrated significant improvements in receptive language skills in pediatric patients aged 10-18 years.
