Overview

donepezil + MK-3134 is a **combination of two small molecules: donepezil**, an acetylcholinesterase inhibitor, and **MK-3134**, a histamine H3 receptor inverse agonist. Donepezil increases acetylcholine concentrations by inhibiting acetylcholinesterase, thus improving cholinergic neurotransmission, while MK-3134 enhances histaminergic neurotransmission by inverse agonism at the H3 receptor. This combination has been studied for synergistic effects on cognitive impairment, particularly in models simulating features of aging and Alzheimer's disease. Clinical trials have investigated the efficacy of this combination for attenuating scopolamine-induced cognitive deficits, supporting the hypothesis that simultaneous modulation of cholinergic and histaminergic pathways could have additive or synergistic benefits for cognition in dementia and related disorders[2][3][6].
