This is a combination of three drugs used in clinical research to study cognitive impairment and its reversal. Donepezil is an acetylcholinesterase inhibitor that increases acetylcholine levels in the brain, commonly used for Alzheimer's disease. MK-3134 is a small molecule inverse agonist of the histamine H3 receptor, developed by Merck Sharp & Dohme, investigated for its potential to enhance cognition by modulating histaminergic neurotransmission. Scopolamine is an anticholinergic agent that induces temporary cognitive impairment and memory deficits, often used experimentally to model aspects of dementia or aging-related cognitive decline. In clinical studies, this combination has been tested to evaluate whether donepezil and/or MK-3134 can attenuate or reverse the cognitive impairments induced by scopolamine administration[1][2][3]. The mechanism involves cholinergic enhancement (donepezil), histaminergic modulation (MK-3134), and cholinergic blockade (scopolamine).
