Overview

This is an investigational donor-derived CD19-targeted chimeric antigen receptor (CAR) T-cell therapy developed by Beijing GoBroad Hospital for the treatment of relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL). The therapy utilizes T cells collected from the patient's original hematopoietic stem cell donor, which are then genetically engineered using a lentiviral vector to express a second-generation CAR construct. This construct typically contains a CD19-binding domain, a 4-1BB co-stimulatory domain, and a CD3ζ signaling domain. This approach is specifically designed for patients who have relapsed after allogeneic hematopoietic stem cell transplantation (allo-HSCT), aiming to induce remission by leveraging donor-matched cells to target B-cell malignancies while managing the risks of graft-versus-host disease (GvHD). In clinical settings, it is often studied as part of a sequential treatment regimen involving CD22-targeted CAR T cells and bridging to or following allo-HSCT.
