Donor-derived CD22 CAR T cells are an investigational allogeneic chimeric antigen receptor (CAR) T-cell therapy developed by Beijing GoBroad Hospital for the treatment of B-cell acute lymphoblastic leukemia (B-ALL). These T cells are harvested from a donor (typically the same donor used for hematopoietic stem cell transplantation) and engineered to express a CAR targeting the CD22 antigen, a protein widely expressed on the surface of B-cell malignancies. In clinical practice, this therapy is utilized as part of a sequential treatment strategy where patients first receive donor-derived CD19 CAR T cells followed by allogeneic hematopoietic stem cell transplantation (allo-HSCT), with the CD22 CAR T cells administered subsequently to consolidate remission and prevent relapse. This approach aims to leverage the graft-versus-leukemia effect while providing a secondary target to overcome potential CD19 antigen escape.
