Overview

donor natural killer cells + ALT-801 is a combination immunotherapy that incorporates two experimental modalities: - Donor-derived (allogeneic) **natural killer (NK) cells**, which are innate immune lymphocytes with the intrinsic ability to recognize and destroy cancer cells; and - **ALT-801**, a bifunctional fusion protein consisting of interleukin-2 (IL-2) linked to a single-chain T cell receptor domain that specifically recognizes a peptide epitope of the human p53 protein presented by HLA-A*0201 on tumor cells. This combination is being developed as a next-generation cancer immunotherapy. It is designed to leverage the cytolytic capacity of NK cells, further potentiated by the immunostimulatory and tumor-targeting effects of ALT-801. ALT-801 activates and directs immune responses predominantly against p53+/HLA-A*0201-expressing tumors, theoretically enhancing recruitment and activation of NK and T cells in the tumor microenvironment and improving anti-tumor activity over standalone IL-2 or NK cell treatments. Clinical investigation is ongoing in oncology indications.
