Overview

This combination cell therapy involves the infusion of purified donor-derived regulatory T cells (Tregs) and conventional T cells (Tcons), typically as part of a high-precision hematopoietic stem cell transplant. Developed primarily by Orca Bio (based on research from Stanford University), the therapy aims to mitigate the risks of allogeneic transplantation. Tregs (CD4+CD25+FOXP3+) are included to suppress the donor-versus-host immune response, thereby preventing graft-versus-host disease (GvHD), which is a major cause of transplant-related morbidity. Tcons (the standard T-cell population) are provided to ensure a robust graft-versus-leukemia (GvL) effect to prevent cancer relapse and to facilitate rapid immune reconstitution against viral and fungal infections. By precisely controlling the ratio and dose of these cell subsets, the therapy seeks to achieve the curative benefits of a transplant while significantly reducing the incidence of severe GvHD without the need for intensive pharmacological immunosuppression.
