Dornase alfa + cisplatin is an investigational combination therapy being developed by the National Cancer Institute in Slovakia for the treatment of relapsed or refractory germ cell tumors. Dornase alfa is a recombinant human deoxyribonuclease I (rhDNase I) that degrades extracellular DNA, including neutrophil extracellular traps (NETs), which have been implicated in promoting chemotherapy resistance and poor prognosis in germ cell tumors. Cisplatin is a standard platinum-based chemotherapy agent that induces cell death by forming DNA adducts and crosslinks. The combination aims to restore cisplatin sensitivity by degrading extracellular DNA in the tumor microenvironment, thereby enhancing the efficacy of the chemotherapy. This approach is currently being evaluated in a Phase II clinical trial (NCT07121322).
