dorzolamide + brimonidine + timolol

This is an investigational triple-combination ophthalmic solution containing dorzolamide hydrochloride, brimonidine tartrate, and timolol, which was developed by Allergan for the treatment of glaucoma and ocular hypertension. The formulation combines three active ingredients with distinct mechanisms of action to lower intraocular pressure (IOP): dorzolamide (a carbonic anhydrase inhibitor), brimonidine (an alpha-2 adrenergic agonist), and timolol (a non-selective beta-blocker). Dorzolamide and timolol primarily work by reducing the production of aqueous humor, while brimonidine both decreases production and enhances uveoscleral outflow. Allergan conducted a Phase 3 clinical trial (NCT01284166) to evaluate the safety and efficacy of this triple therapy in patients who remained uncontrolled on a dual combination of dorzolamide and timolol. Although similar triple-fixed combinations are marketed in certain regions by other companies, Allergan's specific version did not receive regulatory approval or a brand name.
