Overview

Triplet combination therapy of **dostarlimab**, **bevacizumab**, and **niraparib** is an investigational regimen studied in advanced, platinum-resistant ovarian cancer. Dostarlimab is a monoclonal antibody immune checkpoint inhibitor targeting PD-1, enhancing T cell-mediated anti-tumor response. Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), restricting tumor angiogenesis. Niraparib is a small molecule inhibitor of poly(ADP-ribose) polymerase (PARP), preventing DNA repair in cancer cells with homologous recombination deficiencies. The combination aims to exploit synergistic effects in DNA damage response, immune checkpoint blockade, and anti-angiogenic therapy for improved antitumor activity in heavily pretreated ovarian cancer. Developed and studied primarily for platinum-resistant ovarian cancer, this regimen has demonstrated modest activity in clinical trials, with manageable toxicity profiles consistent with individual agents[1][4][7].
