**Dostarlimab + cobolimab** is a combination of two investigational monoclonal antibodies targeting immune checkpoint pathways for cancer immunotherapy. Dostarlimab is an anti–PD-1 monoclonal antibody that blocks the PD-1 receptor, preventing interaction with its ligands PD-L1 and PD-L2 and thereby enhancing T-cell-mediated anti-tumor activity. Cobolimab is an anti–TIM-3 monoclonal antibody that inhibits TIM-3, an immune checkpoint receptor involved in the negative regulation of CD8+ T cells and antigen-presenting cells. The combination seeks to overcome resistance to PD-1 inhibition by dual blockade of PD-1/PD-L1 and TIM-3 pathways, restoring adaptive immune responses against tumors. Clinical trials have shown this combination improves major pathological response rates in high-risk, resectable melanoma and is being investigated in pediatric and adult solid tumor indications[1][3][4][5][7].
