Overview

dostarlimab + GSK4381562 is an investigational combination of two immune checkpoint inhibitors developed for the treatment of cancer. Dostarlimab is a humanized monoclonal antibody that targets the programmed cell death protein 1 (**PD-1**) receptor, thereby blocking its interaction with PD-L1 and PD-L2 and enhancing T-cell mediated anti-tumor response. GSK4381562 is a monoclonal antibody that targets the poliovirus receptor related immunoglobulin domain containing (**PVRIG**), a novel immune checkpoint that modulates immune cell activation in the tumor microenvironment. Early evidence suggests that inhibition of PVRIG by GSK4381562 may enhance recruitment and activation of natural killer cells and further promote anti-tumor activity. This combination is being studied to evaluate safety and efficacy in advanced cancers, including clear cell renal cell carcinoma and other solid tumors, in a phase 1 clinical trial[1][3].
