Dovitinib + fulvestrant is a combination therapy that pairs dovitinib, a potent inhibitor of fibroblast growth factor receptor (FGFR), vascular endothelial growth factor receptor (VEGFR), and platelet-derived growth factor receptor (PDGFR), with fulvestrant, a selective estrogen receptor antagonist. This combination was investigated for treating hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) advanced breast cancer that had progressed during or after prior endocrine therapy. The combination showed promising clinical activity specifically in patients with FGF pathway amplification, with a median progression-free survival of 10.9 months versus 5.5 months in the placebo arm for this subgroup.
