doxifluridine + leucovorin

Doxifluridine (dFUR) is an oral prodrug of fluorouracil (5-FU), a pyrimidine analog antimetabolite, while leucovorin (folinic acid) is a reduced folate that enhances the cytotoxic effects of fluorouracil by stabilizing the ternary complex between thymidylate synthase and 5-fluorodeoxyuridylate. The combination of oral doxifluridine and leucovorin has been studied as an all-oral regimen for advanced colorectal cancer, demonstrating activity in both previously untreated and pretreated patients. The regimen is generally well tolerated, with diarrhea being the main significant side effect[1][2][4].
