Drug intelligence / Profile preview
Lipo-Dox is a pegylated liposomal formulation of doxorubicin hydrochloride developed by TTY Biopharm. It utilizes 'Stealth' technology, where the active anthracycline agent is encapsulated within liposomes coated with polyethylene glycol (PEG). This modification significantly alters the drug's pharmacokinetic profile by extending its circulation half-life and allowing for preferential accumulation in tumor tissues through the enhanced permeability and retention (EPR) effect. Doxorubicin itself acts by intercalating into DNA and inhibiting topoisomerase II, which prevents DNA replication and leads to apoptosis. The liposomal delivery system is designed to reduce systemic toxicities, particularly cardiotoxicity and alopecia, compared to conventional doxorubicin, although it is associated with specific adverse effects such as palmar-plantar erythrodysesthesia. Lipo-Dox is indicated for the treatment of metastatic breast cancer, advanced ovarian cancer, AIDS-related Kaposi's sarcoma, and multiple myeloma.
