Overview

This is a combination chemotherapy regimen composed of five cytotoxic agents: doxorubicin (an anthracycline antibiotic), cyclophosphamide (an alkylating agent), fluorouracil (a pyrimidine analog antimetabolite), paclitaxel and docetaxel (both taxanes). These drugs are used in various combinations for the treatment of breast cancer. Doxorubicin intercalates DNA and inhibits topoisomerase II; cyclophosphamide crosslinks DNA; fluorouracil inhibits thymidylate synthase disrupting DNA synthesis; both paclitaxel and docetaxel stabilize microtubules preventing cell division. The regimens AC-T or TAC are commonly used in adjuvant or metastatic breast cancer settings[1][3][8]. The addition of these agents improves response rates but increases toxicity.
