Overview

doxorubicin + cyclophosphamide + gemcitabine is a multi-agent cytotoxic chemotherapy regimen composed of an anthracycline, an oxazaphosphorine alkylating prodrug, and a deoxycytidine nucleoside analog. Doxorubicin intercalates into DNA and inhibits topoisomerase II while also generating free radicals, cyclophosphamide is metabolically activated to DNA-crosslinking phosphoramide mustard, and gemcitabine is phosphorylated intracellularly to active metabolites that inhibit DNA synthesis through incorporation into DNA and inhibition of ribonucleotide reductase. The combination has been investigated as an antineoplastic regimen, particularly in breast cancer settings including neoadjuvant and metastatic disease, but it is not a uniquely branded fixed-dose product and is best understood as a regimen assembled from its component agents.
