Overview

This is a combination regimen consisting of three drugs—doxorubicin, docetaxel, and tamoxifen—used primarily in the treatment of breast cancer. Doxorubicin is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, leading to DNA damage and apoptosis. Docetaxel is a taxane-class chemotherapeutic agent that stabilizes microtubules, preventing cell division and inducing apoptosis. Tamoxifen is a selective estrogen receptor modulator (SERM) that acts as an antagonist on estrogen receptors in breast tissue, inhibiting the growth of hormone receptor-positive tumors. This combination leverages both cytotoxic chemotherapy (doxorubicin and docetaxel) for broad antineoplastic activity and hormonal therapy (tamoxifen) for targeting hormone-sensitive cancer cells[2][6][7]. The regimen may be considered for locally advanced or metastatic breast cancer, particularly in cases where both cytotoxic chemotherapy and endocrine therapy are indicated.
