Overview

A three-drug antineoplastic regimen combining the anthracycline doxorubicin, the taxane docetaxel, and the anti-HER2 monoclonal antibody trastuzumab. It is used in research and some clinical settings for HER2-positive breast cancer, particularly in neoadjuvant or metastatic contexts. Mechanistically: doxorubicin intercalates DNA and inhibits topoisomerase II, generating DNA damage and free radicals; docetaxel stabilizes microtubules, preventing cell division; trastuzumab targets HER2 (ERBB2), inhibiting downstream signaling, mediating antibody-dependent cellular cytotoxicity, and promoting HER2 internalization. Concurrent use of conventional anthracyclines with trastuzumab has notable cardiotoxicity risk; some studies substitute pegylated liposomal doxorubicin to mitigate this risk.
