Overview

Doxorubicin + epirubicin is a non-standard, investigational combination of two anthracycline antineoplastic antibiotics. Both agents are widely used individually in chemotherapy regimens for various cancers, particularly breast cancer. Doxorubicin and epirubicin act primarily by intercalating into DNA, inhibiting topoisomerase II activity, blocking DNA and RNA synthesis, generating free radicals that cause cytotoxicity, and ultimately inducing apoptosis in rapidly dividing cells. While both drugs share similar mechanisms of action as anthracyclines, they differ slightly in their pharmacokinetics and toxicity profiles; notably, epirubicin is considered to have reduced cardiotoxicity compared to doxorubicin. The use of both together is not standard clinical practice due to overlapping toxicities but may be explored experimentally or discussed in toxicological studies[2][4][5][6][8].
