Overview

Doxorubicin + low molecular weight hyaluronic acid + aminophenylboronic acid (DOX@LR/oHA-APBA) is a pH-responsive, tumor-targeted nanohybrid formulation developed for the treatment of breast cancer. The formulation utilizes laponite RDS (LR) nanodisks as a drug carrier, where doxorubicin is loaded into the interlayer space via ion exchange. The nanodisks are then assembled with a targeted protection layer composed of oligomeric hyaluronic acid (oHA) conjugated to 3-aminophenylboronic acid (APBA). This system targets CD44 receptors overexpressed on cancer cells via the hyaluronic acid moiety, while the boronic acid groups can interact with sialic acid residues on tumor cell membranes. The nanohybrid exhibits acid-dependent, sustained release of doxorubicin in the acidic tumor microenvironment, enhancing cellular uptake and cytotoxicity against breast cancer cells.
