This is a combination chemotherapy regimen consisting of three cytotoxic agents—doxorubicin, mitomycin, and vincristine—used primarily in the treatment of advanced breast cancer and other malignancies. - **Doxorubicin** is an anthracycline antibiotic that intercalates DNA, inhibits topoisomerase II, and generates free radicals leading to DNA damage and apoptosis. - **Mitomycin** is an alkylating agent that crosslinks DNA strands, inhibiting DNA synthesis and function. - **Vincristine** is a vinca alkaloid that binds to tubulin, inhibiting microtubule formation during cell division (mitosis), resulting in cell cycle arrest at metaphase. The combination exploits different mechanisms of action for synergistic antitumor effects but also increases the risk of cumulative toxicities such as myelosuppression and neuropathy. This regimen has been studied particularly in patients with advanced breast cancer who have failed prior therapies[1].
