A combination chemotherapy regimen consisting of doxorubicin, paclitaxel, and cisplatin. This three-drug regimen has been studied primarily for treating ovarian cancer. The combination works through multiple mechanisms: doxorubicin damages DNA inside cancer cells, paclitaxel disrupts the microtubule structure supporting cancer cells, and cisplatin forms DNA crosslinks that prevent cell division. While showing high antitumor efficacy (up to 89% complete clinical response in some studies), this regimen is associated with significant toxicity including neutropenia, renal toxicity, and severe nausea and vomiting. The maximum tolerated dose of doxorubicin in this combination is 30 mg/m² when used with standard doses of paclitaxel (135 mg/m² over 24 hours) and cisplatin (75 mg/m²).
