Overview

This is a combination therapy regimen used primarily for HER2-positive breast cancer. It combines an anthracycline (doxorubicin), a taxane (paclitaxel), and dual HER2-targeted therapy (pertuzumab and trastuzumab). The combination provides synergistic effects in treating HER2-positive breast cancer. Trastuzumab binds to subdomain IV of the HER2 receptor, while pertuzumab binds to subdomain II, preventing HER2/HER3 heterodimerization and homodimerization. This dual blockade of HER2 signaling, combined with the cytotoxic effects of doxorubicin and paclitaxel, has shown improved efficacy compared to single-agent therapies.
