Overview

Doxorubicin liposomal is a pegylated liposomal formulation of the anthracycline cytotoxic agent doxorubicin, marketed as **Doxil** in the United States and **Caelyx** in other markets. Doxorubicin hydrochloride is encapsulated in PEG-coated liposomes, which prolong circulation by reducing uptake by the mononuclear phagocyte system and alter tissue distribution relative to conventional doxorubicin. The released doxorubicin intercalates with DNA, inhibits nucleic-acid synthesis and topoisomerase II activity, producing DNA damage and tumor-cell death. Doxil is approved for platinum-refractory ovarian cancer, AIDS-related Kaposi sarcoma, and multiple myeloma in combination with bortezomib; it has also been evaluated with MR-guided high-intensity focused ultrasound hyperthermia in relapsed or refractory pediatric and young-adult solid tumors.
