Overview
Doxorubicin liposomal + estramustine is a combination of two chemotherapeutic agents used in oncology, particularly for the treatment of cancers such as prostate cancer. Doxorubicin liposomal is an anthracycline antibiotic encapsulated in pegylated liposomes, which prolongs its circulation time and enhances tumor targeting while reducing toxicity. It works primarily by intercalating into DNA, inhibiting topoisomerase II, and generating free radicals that damage cellular components, leading to apoptosis of rapidly dividing cells[1][3][5]. Estramustine is a conjugate of estradiol and nitrogen mustard; it acts as an alkylating agent causing DNA strand breaks and also binds to estrogen receptors due to its estrogenic component[6]. The combination may be considered for hormone-refractory or advanced prostate cancer when other treatments have failed or are unsuitable[8].
