Doxycycline + zinc is a combination therapy evaluated for the primary prevention of COVID-19 infection. Doxycycline is a tetracycline-class antibiotic that exhibits broad-spectrum antibacterial activity and significant anti-inflammatory properties, primarily through the inhibition of matrix metalloproteinases (MMPs). In the context of viral infections, it has been hypothesized to interfere with viral entry and replication. Zinc is an essential trace element that plays a critical role in immune function and has been shown to inhibit viral RNA-dependent RNA polymerase (RdRp) activity in vitro. This specific combination, consisting of 100 mg of doxycycline and 15 mg of zinc daily, was investigated in the Phase 3 'OD-doxy-PNV-COVID-19' trial (NCT04584567) led by Hédi Gharsallah at the Military Hospital of Tunis. The study focused on healthcare workers in Tunisia and demonstrated a significant reduction in SARS-CoV-2 infection rates among participants receiving the combination compared to placebo or doxycycline alone.
