Overview
BON-001 is a delayed-release oral combination tablet containing doxylamine succinate (a first-generation antihistamine) and pyridoxine hydrochloride (vitamin B6). It is being developed by Mochida Pharmaceutical, under license from Duchesnay, for the treatment of nausea and vomiting of pregnancy (NVP) in Japan. Doxylamine succinate acts as a competitive antagonist at histamine H1 receptors and also exhibits anticholinergic activity via muscarinic receptor antagonism, which collectively helps suppress the vomiting center in the brainstem. Pyridoxine hydrochloride serves as a precursor to pyridoxal 5'-phosphate, an essential coenzyme involved in various metabolic processes, although its precise mechanism in alleviating pregnancy-related nausea remains fully elucidated. BON-001 is currently in Phase III clinical development in Japan.
