Overview

Dronabinol + ondansetron is a combination therapy used primarily for the prevention and treatment of chemotherapy-induced nausea and vomiting (CINV). Dronabinol is a synthetic form of delta-9-tetrahydrocannabinol (THC), the principal psychoactive component of cannabis. It acts as an agonist at cannabinoid receptors (CB1 and CB2), exerting antiemetic effects by modulating neurotransmitter release in the central nervous system[4][6]. Ondansetron is a selective serotonin 5-HT3 receptor antagonist that blocks serotonin both peripherally on vagal nerve terminals and centrally in the chemoreceptor trigger zone, thereby preventing nausea and vomiting associated with chemotherapy or surgery[5]. Clinical studies have shown that both agents are effective individually for CINV; however, their combination does not provide significant additional benefit over monotherapy with either agent alone[1][3][6]. Both drugs are generally well tolerated when used together.
