Overview

Dronabinol + palmitoylethanolamide is a combination drug consisting of dronabinol, a synthetic form of delta-9-tetrahydrocannabinol (THC), and palmitoylethanolamide (PEA), an endogenous fatty acid amide. Dronabinol acts primarily as an agonist at cannabinoid receptor CB1 and CB2, modulating neurotransmitter release in the brain and providing antiemetic, appetite-stimulating, and neuroprotective effects. Palmitoylethanolamide exerts anti-inflammatory and analgesic actions mainly through activation of peroxisome proliferator-activated receptor alpha (PPAR-alpha) and modulation of G-protein-coupled receptor 55 (GPR55). The combination is being developed for neurological conditions such as agitation in Alzheimer's disease, Gilles de la Tourette's syndrome, and sleep apnea syndrome. It is designed to leverage both the central effects of THC/dronabinol on cannabinoid receptors and the peripheral anti-inflammatory properties of PEA[2][5].
