Overview

DS-1040b + enoxaparin is an investigational combination therapy evaluated for the treatment of thromboembolic conditions, specifically **acute submassive pulmonary embolism**. DS-1040b is a novel, small molecule inhibitor of the activated form of thrombin-activatable fibrinolysis inhibitor (TAFIa), designed to enhance fibrinolysis (the breakdown of blood clots) and promote thrombus resolution. Enoxaparin is a well-established low molecular weight heparin anticoagulant that inhibits factor Xa and IIa, reducing new clot formation. In clinical studies, DS-1040b is administered as an intravenous infusion while enoxaparin is given subcutaneously, constituting the combination therapy. Clinical data indicate that while DS-1040b added to enoxaparin appears safe and does not significantly alter enoxaparin pharmacokinetics or increase clinically relevant bleeding, it does not further improve thrombus resolution compared to enoxaparin alone in submassive pulmonary embolism[1][3][5].
