Dual-targeting CAR-NK cells (Beijing Biotech) is an investigational chimeric antigen receptor (CAR) natural killer (NK) cell therapy being developed for the treatment of recurrent or progressive glioblastoma (GBM) and other high-grade gliomas (HGG). This therapy utilizes a biomarker-guided approach where patients' tumors are profiled for the expression of three key antigens: Interleukin-13 receptor subunit alpha-2 (IL13Rα2), Epidermal growth factor receptor (EGFR, including the vIII variant), and B7-H3 (CD276). Based on the individual's antigen profile, a specific dual-targeting CAR-NK construct is selected and administered locoregionally via an intracranial catheter or reservoir. This strategy aims to overcome tumor heterogeneity and prevent antigen escape, a common cause of resistance in single-target CAR therapies. The treatment is typically preceded by lymphodepleting chemotherapy with cyclophosphamide and fludarabine.
