Overview
Duligotuzumab + paclitaxel + carboplatin is an investigational combination regimen consisting of three agents: - **Duligotuzumab**: a dual-action humanized monoclonal antibody that blocks ligand binding to both **epidermal growth factor receptor (EGFR)** and **human epidermal growth factor receptor 3 (HER3)**. It is designed to inhibit EGFR- and HER3-mediated oncogenic signaling pathways. - **Paclitaxel**: a well-established small molecule antineoplastic agent that promotes microtubule polymerization and stabilizes microtubules, thereby inhibiting cell division. - **Carboplatin**: a platinum-based small molecule antineoplastic agent that induces DNA cross-linking and damage, resulting in apoptosis of cancer cells. This triple-drug regimen was studied in a phase Ib clinical trial for the first-line treatment of patients with recurrent or metastatic **squamous cell carcinoma of the head and neck**, typically administered intravenously. The combination aimed to achieve complementary mechanisms of tumor cell destruction through receptor blockade and cytotoxicity. The developer of duligotuzumab is Roche.
