Overview

Duloxetine + acetaminophen is a multimodal analgesic combination therapy being investigated for the management of post-endodontic pain. This combination leverages the synergistic effects of duloxetine, a serotonin-norepinephrine reuptake inhibitor (SNRI), and acetaminophen, a central-acting analgesic and antipyretic. Duloxetine functions by increasing the synaptic concentrations of serotonin and norepinephrine, thereby enhancing descending inhibitory pain pathways. Acetaminophen provides analgesia primarily through the inhibition of cyclooxygenase enzymes and prostaglandin synthesis within the central nervous system. The combination is being evaluated by the Chattogram International Dental College as a preemptive treatment to mitigate both acute and chronic pain associated with dental procedures.
