duloxetine + vortioxetine

Duloxetine + vortioxetine is a combination of two antidepressant medications, each with distinct but complementary mechanisms of action. Duloxetine is a serotonin-norepinephrine reuptake inhibitor (SNRI) that increases levels of serotonin and norepinephrine in the brain by inhibiting their reuptake at synaptic terminals. Vortioxetine is classified as a multimodal antidepressant; it acts primarily as a serotonin reuptake inhibitor and also modulates several serotonin receptors, functioning as an agonist or antagonist at different subtypes. Both drugs are approved for the treatment of major depressive disorder (MDD), and duloxetine is also indicated for generalized anxiety disorder, neuropathic pain, fibromyalgia, and chronic musculoskeletal pain. The combination may be considered in select cases where monotherapy has been insufficient; however, co-administration significantly increases the risk of serotonin syndrome due to additive serotonergic effects[1]. This combination should only be used under close medical supervision.
