Drug intelligence / Profile preview
Dulpatatug + HLX87 is an investigational combination therapy being developed by Shanghai Henlius Biotech for the treatment of HER2-positive breast cancer. The regimen consists of dulpatatug (HLX22), a recombinant humanized monoclonal antibody that targets a novel epitope on subdomain IV of the HER2 extracellular region, and HLX87 (GQ1005), a next-generation HER2-directed antibody-drug conjugate (ADC) licensed from GeneQuantum Healthcare. Dulpatatug acts as a HER2 antagonist and binds to a distinct epitope from trastuzumab, allowing for synergistic HER2 receptor blockade, internalization, and degradation when combined with other HER2-targeted agents. HLX87 consists of an anti-HER2 monoclonal antibody conjugated to a potent topoisomerase I inhibitor payload via a stable, cleavable open-chain linker, designed to deliver strong bystander killing effects with minimized systemic toxicity. The combination is currently being evaluated in Phase 2/3 clinical trials as a first-line treatment for HER2-positive recurrent or metastatic breast cancer and as a neoadjuvant therapy for HER2-positive early or locally advanced breast cancer.
