dupilumab + toripalimab is an investigational combination therapy being evaluated for the treatment of microsatellite stable (MSS) colorectal cancer (CRC) with resectable liver metastases. Toripalimab is a humanized IgG4 monoclonal antibody that acts as a checkpoint inhibitor by binding to the programmed cell death protein 1 (PD-1) receptor, thereby preventing its interaction with ligands PD-L1 and PD-L2 and restoring T-cell mediated anti-tumor activity. Dupilumab is a fully human monoclonal antibody that inhibits interleukin-4 (IL-4) and interleukin-13 (IL-13) signaling by specifically binding to the IL-4 receptor alpha (IL-4Rα) subunit. In the context of MSS CRC, which is typically resistant to PD-1 monotherapy, the addition of dupilumab is intended to modulate the tumor microenvironment by inhibiting Th2-type inflammation and potentially enhancing the efficacy of the PD-1 blockade. This specific regimen is being studied in a Phase 1b/2 trial led by investigator Dan Feng.
