Overview

**Durvalumab + cisplatin** is a combination of a monoclonal antibody (durvalumab) and a platinum-based chemotherapy agent (cisplatin). Durvalumab is an immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1), enhancing the immune system's ability to recognize and destroy cancer cells. Cisplatin induces DNA crosslinking, interfering with DNA replication and transcription, ultimately triggering cancer cell apoptosis. This combination is primarily used in combination regimens (often with gemcitabine) for advanced or metastatic biliary tract cancer and muscle invasive bladder cancer, as well as in extensive-stage small cell lung cancer as part of a triplet regimen. The combination has shown improved efficacy compared to chemotherapy alone in several solid tumor settings, particularly in the context of neoadjuvant and first-line therapy in oncology. Durvalumab is developed by AstraZeneca; cisplatin is a widely used generic chemotherapy developed originally by Bristol Myers Squibb.
