A combination therapy consisting of four components: **durvalumab**, a fully human anti-PD-L1 monoclonal antibody; **monalizumab**, a humanized IgG4 monoclonal antibody targeting NKG2A; **mFOLFOX6**, a chemotherapy regimen comprising oxaliplatin, leucovorin (folinic acid), and fluorouracil (5-FU); and **cetuximab**, a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR). This regimen is used in clinical trials to enhance antitumor immune activity and cytotoxicity, most commonly in the treatment of metastatic or advanced solid tumors—including colorectal cancer. Durvalumab blocks PD-L1, permitting T cell-mediated antitumor effects; monalizumab inhibits NKG2A to relieve inhibition of NK and CD8+ T cells; mFOLFOX6 provides cytotoxic chemotherapy; and cetuximab blocks EGFR signaling, inhibiting cancer cell proliferation and survival[1][2][3][4][5][6][7][8].
