Overview

This is a combination therapy consisting of three drugs: durvalumab (MEDI4736), nab-paclitaxel, and gemcitabine. Durvalumab is a human IgG1κ monoclonal antibody that acts as an immune checkpoint inhibitor by blocking programmed cell death ligand 1 (PD-L1), thereby enhancing T-cell mediated anti-tumor responses. Nab-paclitaxel is an albumin-bound formulation of paclitaxel, a microtubule inhibitor that disrupts cell division. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, leading to apoptosis in rapidly dividing cells. This combination has been used primarily in the treatment of pancreatic cancer and other solid tumors, leveraging both immunotherapy and cytotoxic chemotherapy mechanisms[4][7][8].
