Durvalumab + oleclumab is a combination of two monoclonal antibodies developed for cancer immunotherapy. Durvalumab is an anti-PD-L1 antibody that blocks the interaction between Programmed death-ligand 1 (PD-L1) and its receptors, thereby enhancing the immune system's ability to detect and destroy cancer cells. Oleclumab (MEDI9447) targets Ecto 5'-nucleotidase (CD73), an enzyme involved in generating immunosuppressive adenosine in the tumor microenvironment; by inhibiting CD73, it reduces adenosine-mediated immune suppression and promotes antitumor immunity. This combination has shown encouraging clinical activity in non-small cell lung cancer (NSCLC), particularly as consolidation therapy following chemoradiotherapy for unresectable stage III NSCLC, with improved objective response rates and progression-free survival compared to durvalumab alone[1][3][5]. Both agents are being investigated together in multiple phase II/III trials for various solid tumors[2][4].
