This is a combination regimen consisting of three drugs: **Durvalumab** is a human monoclonal antibody that targets programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80, thereby counteracting tumor immune evasion and enhancing anti-tumor immune responses. **Oxaliplatin** is a platinum-based chemotherapeutic agent that forms DNA crosslinks, inhibiting DNA replication and transcription, leading to cell death. **Capecitabine** is an oral prodrug of 5-fluorouracil (5-FU), which inhibits thymidylate synthase and disrupts DNA synthesis in rapidly dividing cells. This combination has been investigated primarily for advanced gastrointestinal cancers such as biliary tract cancer, gastric cancer, esophageal cancer, and colorectal cancer. The rationale for combining an immune checkpoint inhibitor with chemotherapy is to enhance anti-tumor efficacy by both direct cytotoxic effects on tumor cells and modulation of the tumor microenvironment to improve immune response[1][2][3][5].
