Overview

Durvalumab + ramucirumab is an investigational combination therapy consisting of two monoclonal antibodies: durvalumab, an anti–PD-L1 IgG1 antibody (marketed as Imfinzi), and ramucirumab, an anti–VEGFR2 IgG1 antibody (marketeted as Cyramza). Durvalumab blocks the interaction of PD-L1 with PD-1 and CD80, enhancing T-cell mediated immune responses against tumor cells. Ramucirumab inhibits vascular endothelial growth factor receptor 2 (VEGFR2), thereby blocking angiogenesis required for tumor growth. The combination aims to enhance antitumor activity by simultaneously targeting immune evasion and tumor angiogenesis pathways. It has been studied in phase I trials for advanced/metastatic non-small-cell lung cancer (NSCLC), gastric/gastro-esophageal junction adenocarcinoma, and hepatocellular carcinoma after prior therapies[1][3][7].
