Overview

This combination therapy consists of durvalumab, a human monoclonal antibody targeting programmed death-ligand 1 (PD-L1), and regorafenib, an oral multi-kinase inhibitor. Durvalumab, developed by AstraZeneca, works as an immune checkpoint inhibitor by blocking the interaction of PD-L1 with PD-1 and CD80, thereby restoring T-cell mediated anti-tumor immune responses. Regorafenib, developed by Bayer, inhibits a broad range of kinases involved in tumor angiogenesis (VEGFR1-3, TIE2), oncogenesis (KIT, RET, RAF-1, BRAF), and the tumor microenvironment (PDGFR, FGFR). The combination is currently being evaluated in the VIVA trial, a Phase II study sponsored by IRCCS Ospedale Policlinico San Martino, as an adjuvant treatment for patients with stage IV microsatellite stable (MSS) colorectal cancer who have achieved a 'no evidence of disease' (NED) state through surgery or other local-regional interventions.
