This is a combination regimen consisting of three monoclonal antibodies: - **Durvalumab** is an anti-programmed cell death ligand 1 (PD-L1) antibody that blocks the interaction between PD-L1 and its receptors, thereby enhancing T-cell mediated immune responses against tumor cells. - **Tremelimumab** is an anti-cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4) antibody that promotes immune activation by inhibiting CTLA-4, a negative regulator of T-cell activity. - **Bevacizumab** is an anti–vascular endothelial growth factor A (VEGF-A) antibody that inhibits angiogenesis by blocking VEGF-mediated signaling pathways. The combination aims to synergistically enhance antitumor immunity and inhibit tumor vascularization. It has been investigated in phase II clinical trials for advanced hepatocellular carcinoma (HCC) and biliary tract cancers, with ongoing studies evaluating its efficacy and safety as first-line therapy in these indications[2][4][9]. The regimen leverages the STRIDE protocol (Single Tremelimumab Regular Interval Durvalumab), adding bevacizumab to further target tumor angiogenesis[2][4].
