Overview

This combination comprises **durvalumab** (a human monoclonal antibody targeting PD-L1), **tremelimumab** (a human monoclonal antibody targeting CTLA-4), **carboplatin** (a platinum-based chemotherapy agent), and **etoposide** (a topoisomerase II inhibitor). - Durvalumab blocks the PD-L1 pathway, preventing immune evasion by tumor cells and promoting anti-cancer immune responses. - Tremelimumab blocks CTLA-4, releasing inhibition of T-cell activation to further enhance anti-tumor immunity. - Carboplatin induces DNA crosslinking to prevent cancer cell replication. - Etoposide inhibits topoisomerase II, leading to DNA damage and apoptosis. This combo is used in clinical trials and practice primarily for **metastatic non-small cell lung cancer (NSCLC)** without sensitizing EGFR or ALK mutations, as well as under investigation for other advanced solid tumors[3][4].
