Overview

This is a **four-drug combination therapy** comprised of durvalumab (a PD-L1 monoclonal antibody), tremelimumab (a CTLA-4 monoclonal antibody), nab-paclitaxel (an albumin-bound taxane chemotherapy), and carboplatin (a platinum-based chemotherapy). The regimen is under clinical investigation for the treatment of advanced solid tumors, especially metastatic non-small cell lung cancer (NSCLC), metastatic pancreatic adenocarcinoma, and triple-negative breast cancer, among others. Durvalumab blocks PD-L1 to restore T-cell activity against tumor cells. Tremelimumab targets CTLA-4 to increase T-cell activation. Nab-paclitaxel disrupts microtubule function, inhibiting cell division. Carboplatin forms DNA crosslinks, leading to tumor cell death. Used together, these agents combine immune checkpoint inhibition with cytotoxic chemotherapy for a potentially synergistic antitumor effect[1][3][7].
