Y-90 SIRT-University of Colorado, Denver-colorectal cancer refers to an investigational therapeutic regimen being evaluated in a Phase I clinical trial led by the University of Colorado, Denver. This multi-modal approach is designed for patients with refractory metastatic microsatellite stable (MSS) colorectal cancer with liver metastases. The regimen combines dual immune checkpoint blockade—using durvalumab (an anti-PD-L1 monoclonal antibody) and tremelimumab (an anti-CTLA-4 monoclonal antibody)—with localized radiation therapies. These include Yttrium-90 (Y-90) Selective Internal Radiation Therapy (SIRT), which delivers radioactive microspheres directly to liver tumors via the hepatic artery, and Stereotactic Body Radiation Therapy (SBRT). The combination aims to induce immunogenic cell death through localized radiation, potentially transforming the 'cold' tumor microenvironment of MSS colorectal cancer into one that is more responsive to systemic immunotherapy.
