Overview

E2814 + lecanemab is an investigational **combination therapy** for **Alzheimer's disease**, currently being tested in clinical trials. E2814 is a humanized monoclonal antibody that targets the microtubule-binding region (MTBR) of tau protein, aiming to disrupt tau aggregation and inhibit the progression of neurofibrillary tangles that characterize tauopathies. Lecanemab (also known as BAN2401, marketed as LEQEMBI) is a monoclonal antibody that targets amyloid-beta protofibrils to reduce amyloid plaques in the brain. This combination is designed to address both major pathological hallmarks of Alzheimer’s: amyloid-beta accumulation and tau pathology. Current trials evaluate E2814 in combination with lecanemab to determine whether targeting both tau and amyloid pathways provides additive or synergistic disease-modifying effects, especially in early stages of Alzheimer's disease[1][2][3][4][5][6]. The drugs are administered as E2814 intravenous infusion (every four weeks) and lecanemab as a subcutaneous injection (every week)[1][2].
